Abstract:

樟芝只寄生在牛樟樹腐朽的樹幹內,樟芝是台灣民間經常應用的中草藥,含有三萜類、胺基酸、蛋白質、糖類和多醣體。      本研究包括:(1)測量樟芝多糖體之單醣組成,(2)不同含糖培養基對樟芝醣類化合物合成之影響,(3)利用high-performance anion-exchange Chromatography(HPAEC)對於五種樟芝的多醣作定性分析,(4)以B型肝炎病毒產生的MS-G2 cell Line研究五種樟芝多醣體對抗B型肝炎病毒的作用。      本研究的結論為:(1)添加40g/L濃度的葡萄糖和半乳糖,樟芝生長速度均有明顯的增加,且最快速。添加10-80g/L濃度的鼠李糖、木糖、麥芽糖,對樟芝生長速度無明顯之影響。分析培養60天的樟芝菌絲體,添加葡萄糖可以增加阿拉伯糖和葡萄糖的量,添加蔗糖則提高木蜜醇的合成,添加鼠李糖可以提高鼠李糖在細胞中的合成,添加半乳糖可以提高肌醇和葡萄糖的量,木糖則降低葡萄糖和木蜜醇的合成。(2)樟芝子實體和培養的菌絲體之多糖體不同,分析培養26天的樟芝菌絲體,有中性糖,半乳糖,葡萄糖,甘露醇,半乳糖胺。(3)所有的樟芝菌絲多醣體均有抗B型肝炎抗毒的活性,其中B86在50μg/ml有強烈抑制B型肝炎病毒表面抗原的作用,而且比1000單位/ml的α-干擾素還有效,只有B86及35398兩種有對抗B型肝炎病毒核心抗原的活性,所有樟芝菌絲體均無細胞毒性。
